Abstract

This reaction is affected by the intensity of light, the light’s wavelength, and each pigment apart of the photosynthesis yield better results depending on the wavelength of light.

To test the reaction’s results in varying light intensity, the electron acceptor DPIP was measured (which in returns yields the results of the amount of photosynthesis taking place) in varying light treatments.

Abstract

This reaction is affected by the intensity of light, the light’s wavelength, and each pigment apart of the photosynthesis yield better results depending on the wavelength of light.

To test the reaction’s results in varying light intensity, the electron acceptor DPIP was measured (which in returns yields the results of the amount of photosynthesis taking place) in varying light treatments.

By measuring the absorbance(as it decreased, more DPIP was used (turning from the color blue to clear), more photosynthesis was occurring).

It was observed that the optimal light distance for photosynthesis was 30 cm as 24 cm away from the light source was too close and bright for optimal photosynthetic activity.

While 49 cm was too far from the test tube to catch the light emitted by the lamp. In the light wavelength experiment, the color of the lights was changed (i.e. Red, Blue, Green, White, and Dark) to show which wavelength provides for the fastest photosynthesis.

Spinach leaves when accumulating oxygen (a byproduct of photosynthesis) were allowed to float to the surface as a means for measuring photosynthetic activity.

It was seen that blue light (other than the positive white light control) allowed for best photosynthetic ability as green light is often reflected by chlorophyll (main pigment responsible for photosynthesis).

Lastly, in the absorption spectrum experiment, the pigments, Carotene, Chlorophyll a, Xanthophyll, and Chlorophyll b were tested for absorption at different wavelengths.

It was seen that Chlorophyll b, Carotene, and Xanthophyll have best absorption around wavelength 400 – 500 nm while Chlorophyll a peaks in400- 500 nm and 600 – 700 nm showing that green light is the least optimal light for photosynthesis to occur.

These experiments all portrayed the different factors that could affect photosynthesis: light intensity, wavelength, and absorption spectrum.

Introduction

For the light intensity experiment, the effect on photosynthesis depending on the changing light intensities was tested. Thylakoid membranes inside chloroplasts have pigments with Photosystem I’s optimal light wavelength being 700 nm and Photosystem II’s 680 nm.

The solar energy absorbed by these excite the electrons into a higher energy. By substituting NADPH,the electron carrier for photosynthesis, with DPIP the rate of photosynthesis can be observed. DPIP is originally blue, and when reduced to DPIPH2 it becomes clear.

The more reductions of DPIP, the clearer the solution, the less the absorption, and the more the reaction is taking place. Itwas seen to observe how light intensity from a distance of 24cm, 30cm, and 49 cm, the rate of photosynthesis would occur. One tube was left in the dark for a control.
